چکیده
The effect of the amino acid analog FPA on vaccinia virus replication in HeLa cells was investigated, By varying the dose of FPA or the time after infection at which the analog was added to cultures, it was possible to inhibit differentially the synthesis of viral components in vaccinia-infected cells. When suflicient FPA was added t,o GUItures at the t,ime of infection, the synthesis of viral DNA, NP anbigen, infectious virus, and HA was inhibited. The inhibition of synthesis of LS antigen under all conditions studied was only partial. FPA added to cultures at 6 or more hours post-in feetion did not inhibit production of viral antigens or infectious virus. The inhibition was reversed by P,4 added with the FPA, but PA added at later times was progressively less effective in restoring the ability of inhibited infected cultures to produce virus. The proportion of nuclei incorporating t.hymidine-H3 was decreased in cultures infect.ed with vaecinia virus. This decrease was nut seen in cultures receiving FPB at the t,ime of infection.

Effect of 5-fluoro-2'-deoxyuridine on the Synthesis of Vaccinia Virus.
The effect of FUDR on vaceinia virus replication in Heba cells was investiga~d. The incorporation of deoxyuridine-Ha into viral DNA, as determined by autoradiography, was completely blocked by 10-e M FUDR.. This concentration of the analog prevented production of infect, ious virus above levels present in the cultures after washing at the end of the viral adsorption period. The production of vac...

متن کاملInterferon production in chick-embryo cells. The effect of puromycin and p-fluorophenylalanine.
1. When chick-embryo cells were treated with ultraviolet-inactivated influenza virus (Melbourne strain), interferon was produced after a lag period of about 10hr. 2. The addition of small amounts of either puromycin or p-fluorophenylalanine immediately after the virus inhibited the subsequent production of interferon. Both inhibitors primarily affected protein synthesis, and it is concluded tha...
متن کاملDifferential Effects of Inhibitors on the Steps Leading to the Formation of Sv40 Tumor and Virus Antigens
The effect of DNA antagonists and various antibiotics on steps in the synthesis of SV40 virus in green monkey kidney cells was investigated. Both the early forming tumor (T) antigen, as well as the later synthesized virus (V) antigen, were synthesized in the presence of fluorouracil and iododeoxyuridine. Cytosine arabinoside (and fluorodeoxyuridine in starved cells) prevented synthesis of V ant...
متن کاملDiscriminatory inhibition of protein synthesis in cell-free systems by vaccinia virus transcripts.
The effect of vaccinia virus early transcripts on cellular (globin, HeLa, Chinese hamster ovary) and viral (vaccinia, encephalomyocarditis) mRNA function was studied in reticulocyte and wheat germ cell-free protein-synthesizing systems. Vaccinia virus transcripts of two size classes (8-10 S and 4-7 S), generated in vitro by viral cores, inhibited function of cellular and encephalomyocarditis vi...
